Optimization of L-glutaminase production by Penicillium chrysogenum

Abstract
L-glutaminase (E.C 3.5.1.2) was isolated from Penicillium chrysogenum AUMC 14100 gb, Accession No. MN219732. The enzyme was extracellular as judged from the comparison between extracellular and intracellular activities. Optimization of L-glutaminase was carried out in the light of seven parameters. These parameters were: incubation period, carbon sources, nitrogen sources, pH, temperature, inoculum size and agitation speed. The production of L-glutaminase was investigated throughout 8 days and the optimal incubation period for its production was 6 days. The best carbon and nitrogen sources were glucose and L-glutamine, respectively. The optimum concentrations of glucose and L-glutamine were 2 % w/v and 0.5 % w/v, respectively. The optimum pH and temperature for the enzyme production were 8 and 40°C. The inoculum size at 100 µl (107 spore/ml) and agitation speed at 200 rpm proved to be optimal parameters for the enzyme production.
